Cy5® hydrazide,CY5酰肼染料Cyanine5 hydrazide is a reactive dye for the labeling of aldehydes and ketones, an analog of Cy5® hydrazide.This dye reacts smoothly and nearly quantitatively with various carbonyl groups encountered in biomolecules. Avidin-TRITC/罗丹明标记亲和素几乎所有用于标记的物质均可以同亲和素(AV)或链霉亲合素(SA)结合。小分子的有125I、胶体金、荧光素和化学发光物,大分子物质有酶、抗原或抗体、铁蛋白和荧光蛋白等,其中最常用的是酶、异硫氰酸荧光素(FITC)和胶体金. 亲和素或链霉亲和素均为大分子蛋白,其与酶的标记结合物的制备除可用普通酶标记蛋质分子的直接标记法外.由于其特有的与生物素结合的性能,还可以通过与生物素化酶复合物中的生物素结合,间接地与酶形成结合物。以下举例介绍几种制备亲和素(或链霉亲和素)与常用酶形成标记结合物的方法。 (一)亲和素的标记  1.HRP-亲和素结合物的制备   (1)改良过碘酸钠法:新配制的HRP溶液中加入过碘酸钠(NaIO4),将酶分子上的糖基氧化成醛基;依次用醋酸盐溶液(pH4.5—5)和碳酸盐缓冲液(pH9.0)透析,然后在HRP醛基溶液中滴加亲和素搅拌反应,HRP的醛基即可与亲和素的氨基共价结合;加入(NaBH4)终止反应,再透析、离心去沉淀,上清液即含HRP-AV结合物:   (2)戊二醛法:用新配制的HRP与亲和素溶液充分混匀,加双功能交联剂戊二醛室温反应,后者的二个醛基分别与HRP和亲和素分子中的氨基形成Schiff's,碱使二者连接,经充分透析去掉未反应戊二醛,再离心去沉淀等杂质,收集上清液(含HRP-AV结合物)。  2.亲和素-生物素化HRP复合物的制备:  利用亲和素与生物素间特异的结合,将亲和京素与等体积的生物素化酶(HRP-B)按一定浓度比例混合反应后,亲和素即与HRP-B中的生物素结合,形成亲和素—生物素化HRP复合物(avidin-biotin-peroxidase complex,ABC)。      制备ABC复合物时应注意,亲和素和HRP-B的浓度应控制不高于40μg/ml和10μg/ml,否则将增加非特异反应。  3.亲和素-生物素化碱性磷酸酶(avidin-biotinylated—alkaline phosphatase,ABAP)复合物的制备:将亲和素溶液与生物素化AP按4:l(浓度比)比例混合反应(至少十分钟)后即可制得ABAP复合物
